High Estrogen Raises Women’s PTSD Risk

Research in mice shows high hippocampal estrogen at the time of multiple stressors amplifies memory changes, offering insight into sex differences in trauma response.

High levels of estrogen in the brain’s hippocampus during acute stress may explain why women are approximately twice as likely as men to develop post-traumatic stress disorder, according to research published in Neuron. (1)

UC Irvine-led experiments with mice exposed to multiple simultaneous stressors (mimicking events like disasters) showed that females in high-estrogen phases formed stronger, more generalised and longer-lasting fear memories. The effect was linked to estrogen loosening DNA structure, allowing lasting changes in memory circuits. (2)

Vulnerability depended on hormone levels at the time of stress. Blocking specific estrogen receptors prevented the memory impairments. Lead researcher Tallie Z. Baram noted this could open avenues for sex-specific interventions to reduce PTSD and related dementia risks. (1)
