Neural Circuit Target May Improve PTSD Treatment
Lesion and stimulation studies reveal a medial prefrontal-amygdala-temporal circuit where reduced connectivity heightens PTSD risk, offering a target for future therapies.

Recent neuroimaging and lesion studies have pinpointed a neural circuit involving the medial prefrontal cortex, amygdala, and anterolateral temporal lobe where reduced connectivity associates with higher PTSD likelihood (1). In individuals with penetrating brain injuries, lesions disrupting this circuit lowered PTSD development risk.
Cross-validation in non-lesioned participants confirmed lower connectivity in those with PTSD. Transcranial magnetic stimulation in a subset reduced connectivity in this circuit and alleviated symptoms, suggesting noninvasive neuromodulation as a promising avenue (1).
This builds on neurobiological understanding of fear processing and threat response in PTSD. Opinions vary on specificity, but experts view it as a step toward precision treatments, potentially benefiting those with comorbid anxiety or depression.
